Abstract

An efficient protocol was developed for the invitro establishment of axillary buds from material obtainedfrom mature alder specimens of different genotypes. Thecuttings, obtained from 20 to 30 year-old-trees, were forcedflushed and the shoots were cultured in Woody PlantMedium (WPM) (Lloyd and McCown, 1980) with 2 mg/lbenzyladenine (BA) and 0.5 mg/l indol-3-acetic acid (IAA).The cultures were maintained by sequential subcultureevery 9 weeks on the same mineral medium, but with 0.1mg/l BA and 0.5 mg/l IAA. Transfer to fresh medium every 3weeks, the inclusion of zeatin (Z) in the final cycle andsubstitution of 3% sucrose with 2% glucose improved therates of multiplication and the quality of the shoots. Theshoots obtained at the multiplication stage rooted easily,even without auxin (84.0%), although the presence of indol-3-butiric acid (IBA) in the medium increased the rootingsuccess, especially when glucose 2% was used (93.4%) inplace of sucrose 3% (86.9%). The rooted plants wereacclimatized in a growth chamber for 6 weeks before beingtransferred to the greenhouse.
